ABSTRACT
BACKGROUND:
The eradication of Helicobacter pylori is a noble goal that would have a major impact on present and future global health issues related to its infection especially in developing countries. This study evaluated the antioxidant properties of extracts/fractions of Artocarpus altilis and its antibacterial activity against Helicobacter pylori.
METHODS:
Samples of leaves, stem bark and root bark were collected and authenticated at the herbarium of the Department of Botany, University of Ibadan. Air-dried coarse-milled samples were extracted with methanol at room temperature. Filtered extracts were concentrated in vacou, dried with rotary evaporator and stored at 4℃. Antibacterial screening and minimum inhibitory concentration (MIC) were determined by agar diffusion and agar dilution method respectively. DPPH radical scavenging assay method evaluated Antioxidant properties. Combination effect was studied using checkerboard assay and evaluated by fractional inhibitory concentration (FIC) index.
RESULTS:
Stem and root bark methanol extracts inhibited the growth of Helicobacter pylori isolates with zones of inhibition ranging from 16.0 to 30.0 mm and MIC of 50 µg/mL - 200 µg/mL. The most active ethylacetate partition had zone of inhibition of 15.0+ 0.5 mm - 20.0 + 0.5 mm and MIC range of 50 -100 µg/mL. The FIC index for the combination study revealed varied outcomes with the different strains tested. Affinity to DPPH of the root and stem bark of Artocarpus altilis was found to be (76.65%, 73.24%, 64.13%, 60.47%, 53.52%) and (61.82%, 57.78%, 52.83%, 47.97%, 44.39%), respectively compared with ascorbic acid (89.02%, 87.38%, 84.97%, 71.26%, 69.55%) at concentrations of 500 µg/mL - 100 µg/mL.
CONCLUSIONS:
The antibacterial and antioxidant activities of Artocarpus altilis suggest therapeutic potential against Helicobacter pylori, although variable combination effects indicate it should not be used concurrently with standard treatments.
