ABSTRACT
BACKGROUND:
Melanoma remains one of the most aggressive skin cancers worldwide, with rising incidence and significant public health implications, particularly in low- and middle-income countries where access to advanced therapeutics is limited. Current treatment regimens are often costly and associated with adverse effects, necessitating the exploration of affordable, safe, and sustainable therapeutic options aligned with the Sustainable Development Goals (SDGs), particularly SDG 3 (Good Health and Well-being).
OBJECTIVES:
This research collectively investigates the therapeutic potential of MAZ-51, a vascular endothelial growth factor receptor inhibitor, in combination with the dietary phytochemical zingerone, with the aim of uncovering novel adjuvant therapies for melanoma. By integrating molecular pharmacology with a public health lens, the studies explore innovative approaches that can lower treatment barriers and promote health equity.
METHODS:
Two complementary studies were conducted. The first Hlophe Y et al, published in Cellular Biochemistry and Function (2024), examined the synergistic potential of MAZ-51 and zingerone in suppressing melanoma growth and signaling pathways. The second Hlophe Y et al , published in Clinical and Experimental Pharmacology and Physiology (2025), investigated the effects of this combination on melanoma cell proliferation, highlighting the role of phytochemicals in enhancing therapeutic efficacy.
RESULTS:
Both studies demonstrate that MAZ-51 and zingerone act synergistically to inhibit melanoma cell survival, proliferation, and angiogenic signaling. Importantly, the incorporation of phytochemicals such as zingerone underscores the potential of accessible, diet-derived compounds to complement conventional cancer therapies.
CONCLUSIONS:
These findings highlight an innovative and sustainable strategy for melanoma therapy, integrating affordable phytochemical adjuvants into existing treatment paradigms. From a public health perspective, this research addresses the urgent need for equitable cancer care, particularly in resource-limited settings, and contributes to the broader global health agenda by aligning with the SDGs.
